Inject the air into the vial

Hold the vial steady and insert the needle, straight down

Holding the vial firmly and steadily on the table surface with one hand, insert the syringe needle straight down through the rubber stopper using your other hand.

Then slowly inject the air from the syringe into the vial.

The next step is to invert vial-and-syringe, and to fill the syringe.
